Infrared Laser Desorption and Electrospray Ionisation of Non‐Covalent Protein Complexes: Generation of Intact, Multiply Charged Species

We present a novel method enabling the infrared laser desorption and electrospray ionisation (ESI) of protein complexes in their native state. Using this method, we demonstrate surprising generation intact, multiply charged ions myoglobin, non-covalent haemoglobin complex, intact immunoglobulin G antibody states. The observation surviving population is characteristic low internal energy build-up experienced during both from solution subsequent ionisation. Compared to conventional nano-ESI, approach yielded slightly lower average charge states suggesting additional maintenance tertiary structure ionisation, more tolerant salts simpler sample purification procedures. This may enable development high-throughput native-MS methods capable analysing composition sequence multiple macromolecular samples per minute.
